Title: Impaired Degradation Followed by Enhanced Recycling of Epidermal Growth Factor Receptor Caused by Hypo-phosphorylation of Tyrosine 1045 in Rbe Cells

چکیده
The manuscript by Gui et al. reports that impaired EGF receptor degradation in the RBE cholangiocarcinoma cell line correlates with a specific reduction in ligand-induced phosphorylation of EGFR Y1045, the recruitment site for c-Cbl. Phosphorylation of receptor tyrosine 1068 is not compromised, so a defect in the kinase activity of the receptor is not to blame. Figure 1 compares the extent of EGFR degradation after EGF addition to the RBE cells and to immortalized cholangiocyte MMNK1 cells. Receptor degradation is clearly compromised in the RBE cells.

Odin (ANKS1A) Modulates EGF Receptor Recycling and Stability
The ANKS1A gene product, also known as Odin, was first identified as a tyrosine-phosphorylated component of the epidermal growth factor receptor network. Here we show that Odin functions as an effector of EGFR recycling. In EGF-stimulated HEK293 cells tyrosine phosphorylation of Odin was induced prior to EGFR internalization and independent of EGFR-to-ERK signaling. Over-expression of Odin incr...
متن کاملMolecular Docking Based on Virtual Screening, Molecular Dynamics and Atoms in Molecules Studies to Identify the Potential Human Epidermal Receptor 2 Intracellular Domain Inhibitors
Human epidermal growth factor receptor 2 (HER2) is a member of the epidermal growth factor receptor family having tyrosine kinase activity. Overexpression of HER2 usually causes malignant transformation of cells and is responsible for the breast cancer. In this work, the virtual screening, molecular docking, quantum mechanics and molecular dynamics methods were employed to study protein–ligand ...
متن کاملMultiple autophosphorylation site mutations of the epidermal growth factor receptor. Analysis of kinase activity and endocytosis.
We have utilized site-directed mutants to study the role of autophosphorylation of the epidermal growth factor (EGF) receptor in the regulation of receptor kinase activity and ligand-induced endocytosis. A single mutation of the major autophosphorylation site, Y1173, and a double mutation of two autophosphorylation sites, Y1173 and Y1148, did not inhibit kinase activity in vivo, using PLC gamma...
متن کاملEGF‑stimulated AKT activation is mediated by EGFR recycling via an early endocytic pathway in a gefitinib‑resistant human lung cancer cell line.
The receptor tyrosine kinase epidermal growth factor receptor (EGFR) and its ligand epidermal growth factor (EGF) are known to play important roles in malignant tumor cells, and the EGFR signaling pathway is one of the most important targets in various tumors, including non-small cell lung cancer (NSCLC). We reported recently that an aberration in certain steps of EGF-stimulated phosphorylated ...
متن کامل2D-QSAR and docking studies of 4-anilinoquinazoline derivatives as epidermal growth factor receptor tyrosine kinase inhibitors
Introduction: Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor derivatives play an important role in the treatment of cancer. We aim to construct 2D-QSAR models using various chemometrics using 4-anilinoquinazoline-containing EGFR TKIs. In addition, the binding profile of these compounds was evaluated using a docking study. Materials and Methods: In this study, 122 compounds of...
